Práce pojednává o problematice analýzy nových, velmi nebezpečných psychotropních látek ze skupiny fentanylů. Teoretická část je zaměřena na farmakologii a metody chemické analýzy těchto látek. Cílem předložené diplomové práce bylo vyvinout metodu pro separaci a kvantifikaci fentanylu a jeho derivátů furanylfentanylu a sufentanilu pomocí vysokoúčinné kapalinové chromatografie s UV/VIS a elektrochemickou detekcí (HPLC-UV/VIS-ECD). Součástí vývoje metody bylo testování meze detekce (LOD) a stanovitelnosti (LOQ). Hodnoty LOD byly pro fentanyl a jeho deriváty v řádu desítek ng/ml pro ECD detekci, zatímco pro UV/VIS detekci byla LOD v řádu stovek ng/ml. Dále se práce zabývá studiem elektrochemických přeměn vybraných psychotropních látek při jejich anodické oxidaci. Produkty elektrolýzy byly analyzovány jednak kapalinovou chromatografií s hmotnostně spektrometrickou detekcí (HPLC-MS), jednak pomocí on-line spojení elektrochemie a hmotnostní spektrometrie (EC-MS). Na základě analýzy kolizních spekter byly navrženy struktury oxidačních produktů. Hlavními produkty elektrochemických přeměn všech vybraných derivátů fentanylu byly jejich N-dealkylované analogy.The Thesis deals with the issue of new and dangerous psychotropic drugs from the group of fentanyls and their analysis. The theoretical section is focused on pharmacology and methods of chemical analysis of the new psychotropic substances. The goal of this thesis was to develop a method providing separation and quantification of fentanyl and its derivatives furanylfentanyl and sufentanil with the aid of high-performace liquid chromatography with UV/VIS and electrochemical detection (HPLC-UV/VIS-ECD). The development of the method included determination of the detection limit (LOD) and the limit of quantification (LOQ). LOD values were in the order of tens of ng/ml for fentanyl and its derivatives for ECD detection. For UV/VIS detection, LOD values were in the order of hundreds of ng/ml. The Thesis also deals with the study of electrochemical transformation of selected psychotropic drugs during anodic oxidation. The electrolysis products were analysed by high-performance liquid chromatography with mass spectrometry detection (HPLC-MS) and on-line coupling of electrochemistry with mass spectrometry (EC-MS). Based on collision spectra analysis, structures of oxidation products were proposed. The main products of the electrochemical transformations of all studied fentanyl derivatives were their N-dealkylated analogs.
