Cyklin-dependentní kinasy jsou klíčové regulační enzymy, intenzivně studované za účelem pochopení fyziologických procesů buňky, ale i klinické léčby nemocí. V praktické části této bakalářské práce byla úspěšně optimalizována exprese a afinitní purifikace rekombinantního lidského komplexu cyklin-dependentní kinasy 2 s cyklinem A. Získaný komplex byl biochemicky studován a byla potvrzena jeho enzymatická aktivita. Součástí studie byla i charakterizace nového inhibitoru s označením LGR4455, u kterého byl potvrzen kompetitivní charakter inhibice a byla stanovena nanomolární inhibiční konstanta. Vazba do aktivního místa kinasy byla potvrzena vyřešením krystalové struktury komplexu s inhibitorem.Cyclin-dependent kinases are key regulatory enzymes, intensively studied to understand the physiological processes of the cell as well as for clinical treatment of diseases. An experimental part of this bachelor thesis was focused on the expression and affinity purification of the recombinant human complex cyclin-dependent kinase 2 with cyclin A. The purified complex was biochemically studied and its enzymatic activity was confirmed and quantified. Inhibition of catalytic activity by the novel compound, LGR4455, was also investigated and ATP-competitive character of inhibition in nanomolar ranges was determined. Finally, the crystal structure of the complex with the inhibitor that binds to the active site was elucidated
