Bakalárska práca je zameraná na charakterizáciu reaktívnych foriem kyslíka (ROS), kľúčových molekúl a radikálov zapojených v signálnych drahách rastlín. Následkom nadmernej produkcie ROS vplyvom pôsobenia teplotného stresu (TS) môže dochádzať k oxidatívnym poškodením bunkových komponentov, ale aj k aktivácii ďalších signálnych dráh spojených s obranou proti oxidatívnemu stresu prostredníctvom enzýmových aj neenzýmových antioxidantov a syntézy heat shock proteínov (Hsp). V experimentálnej časti bol študovaný vplyv TS pri 40, 45 a 50°C pod dobu 2 h na aktiváciu obranných mechanizmov modelovej rastliny Solanum lycopersicum kultivaru Micro-Tom v časovom úseku 2, 3, 4, 6, 8 a 26 h od vystavenia TS. Vplyvom TS pri 45 a 50°C došlo k významnému poklesu obsahu chlorofylu b. Ďalej TS inicioval nárast dvoch hlavných markerov oxidatívneho stresu, a to peroxidu vodíka a lipidovej peroxidácie. V rámci obranných mechanizmov bola zvýšená aj celková antioxidačná kapacita a pomer GSH2/GSSG počas neskoršej fáze TS. U guajakolperoxidázy bol zaznamenaný signifikantný nárast aktivity najmä po 4 h, aktivita superoxiddismutázy a katalázy vzrástla po 4 a 6 h od vystavenia TS. V ranej fáze boli detekované metódou Western blot vyššie hladíny Hsp70.The bachelor thesis is focused on the characterization of reactive oxygen species (ROS), the key molecules and radicals involved in the plant signaling pathways. As a result of excessive production of ROS due to heat stress (TS), oxidative damage of cellular components can occur. At the same time proceeds activation of other signaling pathways associated with protection against oxidative stress through enzyme and non-enzyme antioxidants and heat shock protein (Hsp) synthesis. In the experimental part the influence of TS at 40, 45 and 50 °C for 2 h was studied on the activation of the defensive mechanisms of Solanum lycopersicum cultivar Micro-Tom in the 2, 3, 4, 6, 8 and 26 h period after exposure to TS. The effect of TS at 45 and 50 ° C showed a significant decrease in chlorophyll b content. Furthermore, TS initiated an increase in two major markers of oxidative stress, namely hydrogen peroxide and lipid peroxidation. Within the defensive mechanisms, the overall antioxidant capacity and the GSH2/GSSG ratio were also increased during the later phase of TS. Guajacol peroxidase showed a significant increase in activity, especially after 4 h, the activity of superoxide dismutase and catalase increased 4 and 6 h after TS exposure. Using Western blot, in the early phase were detected higher levels of Hsp70.
