Kofein je významnou složkou lidské stravy, a přestože má řadu prospěšných účinků na zdraví, je spojován také s určitými zdravotními riziky, jako je například riziko vzniku osteoporózy a podobných onemocnění. Jedním z významných regulátorů kostního metabolismu je receptor pro vitamín D. Tato práce se zabývá vlivem kofeinu na expresi cílových genů tohoto receptoru, které potenciálně mohou souviset s vznikem takových onemocnění. Studován byl vliv kofeinu o koncentraci 0,1; 1; 10 a 100 umol/l v koinkubaci s ligandem receptoru, kalcitriolem, o koncentraci 75 nebo 1 nmol/l v buněčných liniích LS180 a HOS a v lidských osteoblastech. Exprese byla stanovována na úrovni RNA pomocí Real-Time PCR a na úrovni proteinu pomocí Western blottingu, mezi sledovanými geny byly geny související s metabolismem kalcitriolu (CYP24A1 a CYP27B1) a s tvorbou kostní matrix (BGLAP, SPP1 a TNFSF11). V buněčné linii LS180 nebyly zaznamenány změny v expresi genů. V buněčné linii HOS a v kultuře osteoblastů byly zaznamenány mírné modulační účinky na expresi genů, které vyžadují další zkoumání.Caffeine is an important part of human diet. Even though it has a number of beneficial health effects, it is associated with certain health hazards such as the development of osteoporosis and similar diseases. Vitamin D receptor is one of the major regulators of bone metabolism. This thesis is focused on the effect of caffeine on the receptor's target genes that potentially may be related to the development of such diseases. The effect of caffeine was studied at concentrations of 0,1; 1; 10 and 100 umol/l in coincubation with the ligand, calcitriol, at a concentration of either 75 or 1 nmol/l in LS180 and HOS cell lines and human osteoblasts. The expression was assayed at RNA level by Real-Time PCR and at protein level by western blotting. The selected genes were associated with calcitriol metabolism (CYP24A1 and CYP27B1) and with bone matrix formation (BGLAP, SPP1 and TNFSF11). There were no changes in gene expression in the LS180 cell line. In the HOS cell line and osteoblast culture there were some modulatory effects detected but they need further analysis.
