Bakalárska práca sa zaoberá problematikou stanovenia prítomnosti irisínu v krvnom sére. Irisín bol pôvodne rozpoznaný ako hormón podobný myokynínom. Prvá úloha pripisovaná irisinu bola jeho schopnosť indukovať trans-diferenciáciu bieleho tukového tkaniva na hnedé, ktoré funguje ako spaľovač tukov. Bolo preukázané, že irisín môže slúžiť k zavedeniu nových liečebných metód, predovšetkým u metabolických ochorení ako napríklad diabetes typu 2, alebo môže byť využitý pri terapii proti obezite. Cieľom bakalárskej práce bolo zavedenie proteomickej metódy, ktorá umožňuje detekciu prítomnosti irisínu v sére a zároveň jeho citlivú kvantifikáciu pomocou hmotnostnej spektrometrie. V rámci práce boli vybrané tri postupy, 1D SDS-PAGE elektroforéza, precipitácia proteínov acetonitrilom a gélova preparatívna elektroforéza, ktorými boli vzorky séra spracované a zároveň cielené na detekciu irisínu. Žiadna z použitých metód sa v tejto bakalárskej práci nepreukázala ako dostatočne citlivá k stanoveniu prítomnosti irisínu v sére.This bachelor thesis deals with the determination of the presence of irisin in blood serum. Irisin was originally recognized as a hormone-like myokine. The first task attributed to irisin was its ability to induce trans-differentiation of white adipose tissue to brown adipose tissue, which acts as a fat burner. It has been established that irisin can serve to introduction of new terapeutic methods, especially metabolic diseases such as type 2 diabetes, or may be used in the treatment of obesity. The aim of this thesis was the introduction of proteomic method, which allows detection and sensitive quantification of the presence irisin levels by tandem mass spectrometry. There were selected three procedures, 1D SDS-PAGE electrophoresis, precipitation of proteins by acetonitrile and gel preparative electrophoresis, which serum samples were processed and also targeted for irisin detection. None of the methods used in this thesis has not shown to be sufficiently sensitive to determine the presence of irisin in serum.
