Elicitiny jsou malé extracelulární proteiny sekretované patogenními organismy rodů Phytophthora a Pythium, které během interakce s hostitelskou buňkou aktivují obranné reakce rostlin související s produkcí oxidu dusnatého (NO). V rostlinných obranných mechanismech se významně uplatňuje S-nitrosylace cysteinových thiolů, posttranslační modifikace proteinů představující hlavní signální dráhu NO. Na enzymové regulaci hladiny S-nitrosylovaných proteinů se u rostlin podílí S-nitrosoglutathionreduktasa (GSNOR) a thioredoxinreduktasa (TrxR). Tato diplomová práce se zabývá studiem možného vlivu elicitinů kryptogeinu a infestinu a jejich mutantů v poloze 13 a 84 na proces S-nitrosylace u čtyř genotypů Solanum spp. vedoucí k možným změnám rezistence vůči patogenu Pseudoidium neolycopersici. Cílem práce bylo zjistit, jak aplikace elicitinů a jejich mutantů ovlivní hladiny proteinových S-nitrosothiolů, aktivity enzymů GSNOR a TrxR v listech neinfikovaných vzorků a také během patogenese P. neolycopersici. Vlivem studovaných elicitinů došlo k nárůstu proteinových S-nitrosothiolů u neinfikovaných vzorků u všech testovaných genotypů Solanum spp. Změny o různé intenzitě byly zaznamenány v aktivitách enzymů GSNOR a TrxR v závislosti na typu elicitinu a studovaném genotypu.Elicitins are extracellular elicitors secreted by pathogenic organisms of genus Phytophthora and Pythium. Elicitins activate defense responses associated with biosynthesis of nitric oxide (NO) during the plant-pathogen interaction. S-nitrosylation of cysteine residues in proteins, posttranslational modification representing the major NO signaling pathway, plays an important role in the plant defense mechanisms. Two denitrosylation enzymes, S-nitrosoglutathione reductase (GSNOR) and thioredoxine reductase (TrxR), are involved in the regulation of S-nitrosylated proteins levels. The aim of the diploma thesis was to study the influence of elicitins and their mutants on the level of protein S-nitrosothiols in leaves of four Solanum spp. genotypes and during pathogenesis of tomato powdery mildew. The activities of two enzymes involved in denitrosylation processes, GSNOR and TrxR, were analyzed. The studied elicitins caused increased levels of protein S-nitrosothiols in all studied Solanum spp. genotypes. Different modulation of GSNOR and TrxR activities depending on the type of elicitin and the studied genotype was detected.
