Pacienti s diagnostikovaným hepatocelulárním karcinomem mají zpravidla těžkou prognózu, protože běžná terapie ještě nedosahuje příliš pozitivních výsledků. U pacientů s hepatocelulárním karcinomem byly prokázány časté deregulace v cyklin-dependentních kinasach, a proto předmětem této práce bylo otestování čtyř nových inhibitorů cyklin-dependentních kinas na buněčných liniích HepG2 a PRF/PLC/5 a zjištění jejich účinku na maligní buňky. Všechny testované látky indukovaly u rakovinných buněk apoptózu a byl prokázán blok v G2/M fázích buněčného cyklu. Tyto efekty jsou v souladu s inhibicí cyklin-dependentních kinas na buněčné úrovni, které byly navíc prokázány imunodetekcí příslušných substrátů. Prezentované výsledky naznačují, že testované látky v některých svých vlastnostech překonávají jiné vyvíjené inhibitory a mohly by v budoucnu znamenat příslib v možnostech léčby pokročilého hepatocelulárního karcinomu nebo i dalších malignit.Patients with hepatocellular carcinoma have bad prognosis, because the regular treatment has not reached for sufficient results yet. Deregulations in cyclin-dependent kinases are common phenomenon in patiens with hepatocellular carcinoma and due to this reason, appropriate inhibitors are still developing. In this thesis we describe biological characterization of four new inhibitors of cyclin-dependent kinases on cell lines HepG2 and PLC/PRF/5. All tested compounds induced apoptosis in cancer cells and blocked G2/M phases of the cell cycle. These effects are in accordance with the inhibition of cyclin-dependent kinases in cells that have been further proven by immunodetection of the CDK substrates. We showed that tested inhibitors overcame biological effects of previously described CDK inhibitors and therefore their further testing could help to succesfull treatment of patients with advanced stage of hepatocellular carcinoma or other malignancies in the future.
