Předložená diplomová práce se zaměřuje na vývoj rychlé chromatografické separace syntetických barviv a jejich analýzu v potravinových vzorcích při odhalování pančování potravin. Barviva mohou být nepovoleně použita pro vylepšování vzhledu potravin. Přidávána mohou být bez uvedení na obalu nebo ve vyšším než povoleném množství. Úvodní kapitoly jsou zaměřeny na možné způsoby pančování potravin jak v minulosti, tak v současnosti. Dále práce poskytuje přehled možných způsobů analytické kontroly pravosti potravin, pozornost je věnována analýze syntetických barviv chromatografickými metodami. Experimentální část přibližuje analýzu syntetických barviv. Chromatografické dělení pomocí superkritické fluidní chromatografie probíhalo na ultraúčinných kolonách s částicemi náplně menšími než 2 ?m a k selektivní a citlivé detekci byl použit hmotnostní spektrometr. Kvalita separace byla posuzována pro různé experimentální podmínky, konkrétně pro různé stacionární fáze, aditiva, gradienty a průtoky mobilní fáze a teploty kolony. Z výsledků měření vyplývá, že nejlepší volbou je 2-Ethylpyridinová stacionární fáze, strmý gradient modifikátoru (methanol/voda s hydroxidem amonným jako aditivem) v oxidu uhličitém, průtok 0,7 ml/min a teplota kolony 40 °C. Doba samotné separace je velmi příznivá a nepřesahuje čas devíti minut. Finální metoda byla částečně validována v parametrech opakovatelnosti, preciznosti, linearity, meze detekce a kvantifikace, zároveň byla aplikována na reálné vzorky iontového nápoje a žvýkacích bonbónů.The presented diploma thesis focuses on the development of fast chromatographic separation of synthetic dyes and their analysis in food samples to control food adulteration. Synthetic dyes can be added illegally to food to improve the appearance. They can be added without any note on food packaging or their content can be higher than accepted limit. The introductory chapters are focused on food adulteration both in the past and nowadays. Further, the work provides an overview of analytical control of food authenticity and the attention is paid to the determination of synthetic dyes by chromatographic methods. The experimental part describes the analysis of synthetic dyes. Chromatographic separation by supercritical fluid chromatography was performed on ultra-efficient columns with stationary phase particles with diameter less than 2 m. A mass spectrometer was used for selective and sensitive detection. The separation was assessed for various experimental conditions, namely for different stationary phases, additives, gradients and flow-rates of mobile phase and column temperatures. Results show that the best choice is a 2-Ethylpyridine stationary phase, a steep gradient of modifier (methanol/water with ammonium hydroxide as an additive) in carbon dioxide, a flow rate of 0.7 ml/min and column temperature of 40 °C. Separation time is very favorable and does not exceed nine minutes. The final method was partially validated in repeatability, precision, linearity, limits of detection and limits of quantification, and was applied to real samples of ionic beverage and chewy candies.
