This bachelor thesis deals with influence of graphene oxide nano-platelets on the transcriptional activity of the nuclear receptor for vitamin D (VDR). The theoretical part of this thesis defines the current statement of this problematics. Vitamin D functions are described, also its isoforms and its importance for human organism. The next chapter is dedicated to the family of nuclear receptors and we can find here also more information about the studied nuclear receptor, the vitamin D receptor. The following chapter deals with nanomaterials, graphene and graphene oxide itself, its structure and significance. The experimental part is divided into two sets of experiments. In the first part, the cytotoxic effect of the graphene oxide nano-platelets on the VDR-responsive cell line was detected by the MTT test. The results demonstrated that none of used concentrations of graphene oxide had toxic effect on the VDR-responsive cell line and therefore all tested concentrations could be used for all other experiments. In the second part, it was determined the ability of these particles to activate or inhibit the transcriptional activity of VDR using the Gene reporter assay. VDR activity was determined using a reporter gene for luciferase, which is under the control of VDR responsive elements.
