Tato rigorózní práce se zabývá validací metod pro stanovení elementárních nečistot v aktivních farmaceutických substancích, meziproduktech a výchozích surovinách podle nových kapitol amerického lékopisu pomocí hmotnostní spektrometrie s indukčně vázaným plazmatem. V teoretické části práce je uveden popis základního principu a instrumentace ICP-MS a podán přehled o nových lékopisných kapitolách. Experimentální část práce je zaměřena na validaci metod ICP-MS podle požadavků USP 232Elemental impurities - Limits a USP 233Elemental impurities - Procedures a souvisejících dokumentů. V rámci provedených validací byly stanovovány meze detekce a kvantifikace, linearita přesnost, správnost, selektivita a robustnost metody. Analyzováno bylo celkem 20 prvků (Cd, Pb, As, Hg, Ir, Os, Pd, Pt, Rh, Ru, Cr, Mo, Ni, V, Cu, Ag, Co, Tl, Se, Au). S výjimkou osmia byla pro všechny prvky splněna všechna předepsaná kritéria. Testování bylo provedeno na dvou aktivních farmaceutických substancích a třech finálních farmaceutických produktech s orálním, parenterálním a inhalačním podáním.This rigorous thesis is focused on the validation of methods for the determination of elementary impurities in active pharmaceutical substances, their intermediates, and starting materials in accordance with novel USP General Chapters by inductively coupled plasma mass spectrometry (ICP-MS). The theoretical part of the thesis describes basic ICP-MS principles and instrumentation and USP General Chapters. The experimental part of the thesis deals with the validation of ICP-MS methods according to USP 232Elemental impurities - Limits and USP 233Elemental impurities - Procedures and related documents. For ICP-MS method, the limit of detection, the limit of quantification, linearity, accuracy, precision, specificity and robustness were established. In total, 20 different elements (Cd, Pb, As, Hg, Ir, Os, Pd, Pt, Rh, Ru, Cr, Mo, Ni, V, Cu, Ag, Co, Tl, Se, Au) were analyzed. With the exception of osmium, all of analyzed elements accomplished required criteria. Altogether, two active pharmaceutical substances and three final pharmaceutical products in oral, parenteral and inhalation forms were tested.
