Cílem práce bylo sledování elektrochemické oxidace sildenafilu citrátu, využití redoxních reakcí pro stanovení analytu a identifikace vznikajících oxidačních produktů pomocí hmotnostní spektrometrie. Elektrochemická oxidace sildenafilu o koncentraci 1x10-4 mol/l byla sledována cyklickou a diferenčně pulzní voltametrií v Britton-Robinsonově pufru o pH 2,00 v přítomnosti 10 % (v/v) acetonitrilu. Mez detekce sildenafilu byla 9,6x10-7 mol/l. Vznikající N-desmethylsildenafil a další oxidační produkty byly detekovány v on-line spojení elektrochemie s hmotnostní spektrometrií.The aim of this work was to study the electrochemical oxidation of sildenafil citrate, apply the redox reactions for analyte determination and investigate the formed oxidation products. The electrochemical oxidation of sildenafil citrate at concentration 1x10-4 mol/l was performed by cyclic voltammetry and differential pulse voltammetry in solution of Britton-Robinson buffer with pH = 2.00 and in the presence of 10 % (v/v) acetonitrile. Limit of detection was 9.6x10-7 mol/l. Formed N-desmethylsildenafil and other oxidation products were detected by on-line coupling of electrochemistry with mass spectrometry.
