Bakalářská práce se zabývá problematikou rekombinantních molekul DNA od etap vývoje této technologie přes její aplikaci při tvorbě biologických léčiv. Součástí této práce je i přehled širokého využití biologických léčiv v praxi. Tato práce také zasahuje do oblasti schvalování léčiv včetně demonstrace rozdílů schválení biosimilars (follow on biologics) a generických léčiv. V neposlední řadě se práce zabývá základními přístupy procesu tvorby rekombinantní DNA. Experimentální část je zaměřena na problematiku lymské boreliózy, konkrétně na purifikaci povrchových antigenů OspC a OspA Borrelia burgdorferi s odstraněnou lipidizační sekvencí na N-terminálním konci. Oba proteiny byly úspěšně purifikovány a výsledky této práce přispěly k optimalizaci podmínek purifikace těchto proteinů. Ústav imunologie Univerzity Palackého navazuje na tyto výsledky s cílem potenciální možnosti vývoje vakcíny jak pro účely veterinární, tak následně i humánní medicíny.The thesis deals with the issue of recombinant DNA molecules from the stages of the development of this technology through its application in the biological drugs production. A part of this thesis involves an overview of the wide use of biological drugs in practice. The thesis, however, focuses also on the area of drug approval, including a demonstration of differences in the approval of biosimilars (follow on biologics) and generic pharmaceuticals. Last but not least, the thesis discusses the basic process approaches of the recombinant DNA production. The experimental part focuses on the issue of Lyme borreliosis, specifically on the purification of surface antigens OspC and OspA of Borrelia burgdorferi including the removed lipid sequencing to the N-terminal end. Both proteins were successfully purified and results of this thesis have contributed to the optimization of purification conditions of these proteins. Moreover, the Department of Immunology at Palacky University followed up on these results with an aim of a potential possibility for development of a vaccine both for veterinary purposes and, consequently, human medicine.
