Methylace DNA je epigenetická modifikace, ke které nejčastěji dochází na pátém uhlíku v cyklické struktuře cytosinu. Tato kovalentní úprava DNA je esenciální pro správný vývoj organismu a je důležitou součásti procesu inaktivace chromozomu X, imprintingu a regulace exprese genetické informace. V této bakalářské práci je popsána stručná charakteristika methylace DNA, její vznik a vliv na expresi genů. Dále jsou v této práci zmíněny a popsány příklady epigeneticky modifikovaných genů, které mají významný podíl na vzniku rakovinných onemocnění. Vzhledem k faktu, že methylační profil buněk lze detekovat pomocí sekvenačních technik, jsou v dalších kapitolách popsány principy a využití sekvenační metody nové generace používané na platformě Illumina a dále jsou vysvětleny vybrané metody detekce methylace DNA a formáty souborů pro analýzu sekvenačních dat. Experimentální část této práce se zaměřuje na celogenomovou detekci methylačních profilů DNA pomocí metody Methyl-Capture sekvenace na platformě Illumina a následném zpracování výstupních dat. Hlavním cílem práce bylo detekovat a identifikovat methylované oblasti v genomech buněčné linie HCT116 odvozené od kolorektálního karcinomu, které mohou mít určitou roli v navozování rezistence buněk vůči demethylační látce decitabinu.DNA methylation is an epigenetic modification, which mainly occurs at the fifth position of carbon in the cyclic structure of cytosine. This covalent modification is essential for regular development of organisms and is an important part of processes such as X-chromosome inactivation, imprinting and regulation of gene expression. In this thesis there are described basic characteristics of DNA methylation, its formation and effect on gene expression. Further, there are mentioned and described examples of epigenetically modified genes, which have significant role in carcinogenesis. Due to the fact that methylation profile of cells can be detected by sequencing techniques, in the next chapters there is also described method of next generation sequencing used on the platform produced by Illumina, several methods of detection of DNA methylation and chosen format files used for analysing sequencing data. The experimental part of this work is focused on the whole-genome detection of DNA methylation profiles by Methyl-Capture sequencing on platform created by Illumina and data processing. The main object of this work was to detect and identify methylated regions in genomes of the HCT116 cell line derivated from cancerous colorectal cells, which can have an important role in inducing resistance to demethylating drug decitabine.
