Inhibice cyklin-dependentních kinas (CDK) je považována za vhodnou strategii léčby rakoviny. V průběhu posledních 25 let byla vyvinuta celá řada inhibitorů CDK, přičemž některé z nich prochází klinickým testováním pro léčbu různých typů rakovin a palbociclib (PD-0332991, Ibrance) byl nedávno schválen pro léčbu metastazující rakoviny prsu. V současné době jsou nově vyvíjené inhibitory CDK testovány především vůči CDK1-CDK9 komplexům, kromě toho jsou ale také profilovány vůči širokému panelu dalších kinas pro stanovení možných nežádoucích cílů (tzv. off-targetů) v rámci kinomu. Nicméně méně známí členové CDK rodiny, CDK10-CDK20, mají také důležité biologické funkce a jejich inhibice může vést k nežádoucím vedlejším účinkům, anebo by naopak mohla být pro terapii přínosná. Teoretická část diplomové práce se zabývá důležitými vlastnostmi CDK10-CDK20 a také metodami pro stanovení inhibiční aktivity a selektivity inhibitorů proteinkinas. Praktická část práce je zaměřena na srovnání odlišných metod pro stanovení kinasové inhibiční aktivity s použitím komerčně dostupných CDK inhibitorů.Inhibition of cyclin-dependent kinases (CDKs) has been considered as a strategy for a treatment of cancer. A number of cyclin-dependent kinase inhibitors have been developed during the last 25 years. Some of them have entered clinical trials for the treatment of different types of cancer and palbociclib (PD-0332991, Ibrance) was recently approved for the therapy of metastatic breast cancer. Currently newly developed CDK inhibitors are tested especially against CDK1-CDK9 complexes. In addition, they are also profiled against broader panel of other kinases from kinome to understand their possible off-targets. However, less known members of CDK family, CDK10-CDK20, have been shown to have important biological functions so their inhibition could lead to undesirable side effects or on the contrary it could be beneficial for the therapy. The theoretical part of diploma thesis describes important properties of CDK10-CDK20 and methods for determining of inhibition activity and selectivity of protein kinases inhibitors. The experimental part is focused on the comparing of different methods for determination of kinase inhibition activity with using four commercially available CDK inhibitors.
