Fytopatogenní houby jsou příčinou velké řady onemocnění rostlin. Do této skupiny patří také houby rodu Botrytis, které na napadených rostlinách způsobují převážně plísně, vadnutí a hnilobu. Hlavním cílem této diplomové práce bylo studium proteinového profilu u Botrytis cinerea a Botrytis tulipae, se zaměřením se na struktury pomáhající houbě šíření infekce a přežítí nepříznivých podmínek jako jsou konidie a sklerocie. Ke studiu byly zvoleny dva různé přístupy. Prvním byla rychlá identifikace mikroorganismů měřením intaktních buněk a proteinů prostřednictvím hmotnostní spektrometrie s ionizací za účasti matrice. Jako nejvhodnější matrice pro měření intaktních konidií a jejich proteinů u rodu Botrytis byla vybrána směs kyseliny ferulové a sinapové. Srovnáním hmotnostních spekter dvou druhů hub, Botrytis cinerea a Botrytis fabae, bylo možné tyto organismy stejného rodu rychle odlišit. Dalším využitým přístupem byla celková analýza proteomu konidií a sklerocií hub. Po sběru biologického materiálu a následné extrakci proteinů, bylo využito metod gelové elektroforézy, kapalinové chromatografie a hmotnostní spektrometrie pro identifikaci a charakterizaci proteinů. Pomocí bioinformatických nástrojů pro predikci funkce proteinů bylo zjištěno, že značná část nalezených proteinů je zapojená do metabolických procesů organismu.Phytopathogenic fungi cause a wide range of plant diseases. The fungus Botrytis is widespread phytopathogen, which is responsible for mold, wilt and rot on infected plants. The main aim of this thesis was to study the protein profile of Botrytis cinerea and Botrytis tulipae with focus on konidia and sclerotia, structures playing a role in a spread of infection and survive in adverse conditions. Two different approaches were selected for study of Botrytis proteome. The first was the rapid identification of microorganisms by measurement of intact konidia and proteins by matrix assisted laser ionization mass spectrometry. The mixture of ferulic and sinapinic acid were seleced as the most suitable matrix for the measurement of intact cells and their proteins of Botrytis. The rapid distinction between two fungal species of the same genus, Botrytis cinerea and Botrytis fabae, were done by comparison of the mass spectra. Another approach used in the thesis was the shotgun proteome analysis of fungal konidia and sclerotia. After harvesting the biological material and the subsequent extraction of the proteins, the gel electrophoresis, liquid chromatography and mass spectrometry methods were used for identification and characterization of proteins. With bioinformatics tools to predict the function of proteins was found that the most proteins are involved in the metabolic processes of the organism.
