Mezi životně důležité buněčné struktury se řadí cytoskelet, který představuje síť proteinových vláken lokalizovaných v cytoplazmě. Zajišťuje růst a prodlužování buňky, její dělení a oporu. U rostlin je cytoskelet tvořen dvěma druhy proteinových vláken: mikrotubuly a aktinová filamenta. Stavební složkou mikrotubulů jsou proteiny alfa tubulin a beta tubulin, u aktinových filament je to aktin. Pro studium cytoskeletu je možno použít přírodní toxiny, které interagují s cytoskeletem. V této bakalářské práci byl použit diterpenoidní alkaloid taxol. Taxol se váže na beta tubulin a inhibuje depolymerizaci mikrotubulů, čímž narušuje jejich funkci. Proteomická analýza umožňuje zjistit proteinové složení v buňce, pletivu, orgánu nebo celém organismu. Teoretická část práce je zaměřena na popis rostlinného cytoskeletu s důrazem na mikrotubuly, charakterizaci taxolu, jeho vlivu na růst rostlin a využití pro proteomickou analýzu. V experimentální části je vyhodnocen vliv taxolu na fenotyp kořenů Arabidopsis thaliana, na dynamiku mikrotubulů a dále vyhodnocení proteomické analýzy vlivu taxolu na kořeny Arabidopsis thaliana. Na základě výsledků proteomické analýzy byly vybrány proteiny (Beclin 1, Sec14p a profilin), které vykazovaly změnu abundance po ošetření taxolem. Tyto proteiny mohou zajišťovat propojení cytoskeletu s transportem vezikul a také propojení mikrotubulů a aktinových filament. Jejich změna abundance po ošetření taxolem byla ověřena metodou imunoblotování u profilinu nebo pozorováním změny intenzity fluorescence u Beclin 1 a Sec14p fuzních konstruktů s GFP pod kontrolou nativního promotoru ve stabilně transformovaných semenáčcích Arabidopsis. Metodou imunoblotování byla také analyzována změna abundance alfa tubulinu, beta tubulinu a aktinu po ošetření taxolem v kořenech Arabidopsis ekotyp Columbia a změna abundance těchto proteinů u mutanta Arabidopsis der1-3, který má nefunkční isovariantu aktin2 pro sledování propojení aktinových filament a mikrotubulů. Výsledky práce poukázaly nejen na inhibiční účinek taxolu na růst primárních a postranních kořenů, gravitropismus a na zvýšení abundance proteinů Beclin 1 a Sec14p, ale i na těsné propojení aktinového cytoskeletu a mikrotubulů.The cytoskeleton is a very important cellular structure. It represents a network of protein filaments which are localized in cytoplasm. Cytoskeleton allows cell growth, elongation, division and mechanical support. Plant cytoskeleton consists of two protein filaments: microtubules and actin filaments. Structural components of microtubules are alpha tubulin and beta tubulin. Actin filaments are composed of protein called actin. Natural toxins which interacts with cytoskeleton can be used for cytoskeleton investigation. In this thesis we used diterpenoid alkaloid taxol. Taxol binds to beta tubulin and inhibits microtubule depolymerization, which alters their fuction. Proteomics allows the identification of proteins in cell, tissue, organ or in whole organism. The theoretical part of this thesis focuses on description of plant cytoskeleton, mainly microtubules, characterization of taxol and its effect on plant growth. The current knowledge about the effects of taxol on proteome of organisms is summarised as well. The experimental part shows evaluation of taxol induced changes in Arabidopsis thaliana root proteome, impact of taxol on microtubule dynamics and Arabidopsis root phenotype. For validation of proteomic data, we selected Beclin 1, Sec14p and profilin which showed changes in their abundance after taxol treatment. These proteins are promising candidates to play important role in crosstalk between cytoskeleton and vesicular trafficking, as well as between actin and microtubules. The change in their abundance was verified using immunoblotting (for profilin) or by microscopic observation of stably transformed Arabidopsis plants expressing Beclin 1 and Sec14p-GFP fusion constructs under their native promoter. Moreover, the actin-tubulin interaction was studied by imunoblotting-based monitoring of alfa tubulin, beta tubulin and actin abundance after taxol treatment in Arabidopsis Columbia ecotype and Arabidopsis der1-3 mutant which express nonfunctional actin2 isovariant. Results showed not only that taxol inhibits growth of primary and lateral roots, gravitropism and cause increase of Beclin1 and Sec14p abundance, but also that actin filaments and mictotubules are tightly connected in their functions.
