Diplomová práce se zabývá problematikou vlivu agonistů jaderných receptorů PPAR na růstový potenciál buněčných linií. Teoretická část se převážně věnuje významu jaderných receptorů PPAR, jejich ligandům, zejména fibrátům, a jakou úlohu hrají PPAR v karcinogenezi a chemoprevenci. Experimentální část se zabývá stanovením viability u buněčných linií HEK293 a HeLa po ovlivnění fibráty. Stanovení bylo prováděno na základě WST testu. Dále byla stanovována exprese vybraných antigenů v buněčných liniích HEK 293 a HeLa po ovlivnění fibráty. Stanovení bylo provedeno na základě imunocytochemie.The diploma thesis deals with the effect of PPAR agonists of nuclear receptors on the growth potential of the cell lines. The theoretical part mainly deals with the importance of PPAR nuclear receptors, their ligands, especially fibrates, and role of PPAR in carcinogenesis and chemoprevention. The experimental part focuses on viability in cell lines HEK293 and HeLa, after influencing by fibrates. The assay was performed by using WST assay. Also was focused on the expression of selected antigens in cell lines HEK293 and HeLa, after influencing by fibrates. The assay was performed by using imunocytochemistry.
