Bakalářská práce pojednává o problematice Barrettova jícnu, prekancerózní lézi, jež předchází vzniku adenokarcinomu jícnu, a také o úloze miRNA představujících novou třídu biomarkerů pro včasnou detekci maligního nádoru. V úvodu teoretické části je objasněna definice Barrettova jícnu, dále jsou popsány příznaky a rizikové faktory tohoto onemocnění. Je také vysvětleno, jakým způsobem lze Barrettův jícen diagnostikovat a jsou zmíněny možnosti léčby. Další kapitola se zabývá funkcí miRNA, včetně jejich biogeneze. Popisuje možnosti detekce neznámých miRNA či metody, jimiž lze sledovat profily genové exprese miRNA. Praktická část zahrnuje popis izolace miRNA ze vzorků odebraných od pacientů trpících Barrettovým jícnem pomocí kartáčkové biopsie. Soustředí se na microarray analýzu, díky níž byly stanoveny profily miRNA exprese u pacientů s Barrettovým jícnem.This bachelor thesis deals with the issue of Barrett's oesophagus - a precancerous lesion that prevents oesophageal adenocarcinoma, and also about the role of miRNAs representing a new class of biomarkers for early detection of malignant tumours. The introduction of the thesis illustrates the definition of Barrett's oesophagus, also describes the symptoms and risk factors of this disease. Furthermore, it Barrett's oesophagus diagnostics, and there are also discussed treatment options. Another chapter deals with the functions of miRNA including their biogenesis. It also gives the possibilities of detection unknown miRNA or methods which can be monitored miRNA expression profiles. The practical part involves isolating miRNA from brush biopsy samples taken from patients suffering from Barrett´s oesophagus. It also concentrates on microarray analysis thanks to which miRNA expression profiles were determined in patients with Barrett´s oesophagus.
