S-nitrosylace, reverzibilní navázání NO skupiny na thiolový postranní řetězec cysteinu v proteinech, představuje jednu z důležitých posttranslačních modifikací v rámci buněčné signalizace oxidu dusnatého. Má významnou úlohu v řadě fyziologických i patofyziologických procesech rostlin. Tato práce se zabývá studiem S-nitrosylace v průběhu patogenese Phytophthora infestans u tří genotypů rajčete Solanum lycopersicum cv. Amateur, Solanum chmielewskii, Solanum habrochaites v časech 0, 24, 48, 72 a 96 hodin po inokulaci. Vzorky byly značeny pomocí metody biotin-switch a S-nitrosylované proteiny byly imunochemicky detekovány či purifikovány afinitní chromatografií. Rovněž byl stanoven obsah proteinových S-nitrosothiolů Savillovou metodou a aktivita enzymu S-nitrosoglutathionreduktasy (GSNOR). Inokulace patogenem způsobila lokální nárůst hladiny proteinových S-nitrosothiolů (RSNO) u všech studovaných genotypů zejména v čase 72 hpi. Naopak, došlo k poklesu aktivity GSNOR, klíčového enzymu katabolismu GSNO, který jako donor NO může ovlivňovat hladinu ostatních RSNO. Detekce a následná purifikace proteinových S-nitrosothiolů potvrdila zvýšenou S-nitrosylaci 72 hpi, a to zejména u genotypu S. lycopersicum cv. Amateur.S-Nitrosylation, the reversible attachment of the NO group to the thiol side chain of cysteine, is one of the most important protein post-translational modifications in the nitric oxide cell signaling. It plays an important role in many plant physiological and pathophysiological processes. This work deals with the study of protein S-Nitrosylation during the pathogenesis of Phytophthora infestans in three genotypes of tomato (Solanum lycopersicum cv. Amateur, Solanum chmielewskii, Solanum habrochaites) 0, 24, 48, 72, 96 hours after inoculation. Samples were labeled by the biotin-switch method and S-Nitrosylated proteins were immunochemically detected or were purified by affinity chromatography. The protein S-nitrosothiols content was determined by Saville method and the enzyme activity of S-Nitrosoglutathione reductase (GSNOR) was measured. Pathogen infection caused local increase of the protein S-nitrosothiols (RSNO) level especially at 72 hpi. Conversely, the GSNOR activity was decreased. GSNOR is the key enzyme in the catabolism of GSNO which as the NO donor may influence the level of other RSNO. Detection and subsequent purification of the protein S-nitrosothiols confirmed the increased S-Nitrosylation 72 hpi, especially for genotype S. lycopersicum cv. Amateur.
