Antimikrobiální peptidy jsou důležitou součástí humorální imunity živočichů. Vyskytují se napříč všemi organismy včetně hmyzu. U této skupiny živočichů bylo také dosud izolováno a popsáno největší množství antimikrobiálních peptidů. Tato práce se zabývá základní charakteristikou antimikrobiálních peptidů, jejich rozdělením a mechanismem působení. Je zaměřena na antimikrobiální peptidy včely medonosné (Apis mellifera), jejichž zástupci (apidaecin Ia, abaecin, jellein I, jellein II) jsou také testováni v praktické části. Peptidy jsou testovány vůči bakteriálním kmenům Escherichia coli, Micrococcus luteus a také Paenibacillus larvae, který způsobuje onemocnění mor včelího plodu. Pro testování antimikrobiální aktivity peptidů pomocí difúzních testů byl optimalizován způsob kultivace, testování vhodných médií i aplikace peptidů na bakteriální kultury. Byla zjištěna citlivost bakteriálních kmenů E. coli vůči apidaecinu Ia. Pro stanovení minimální inhibiční koncentrace apidaecinu Ia byla použita metoda měření růstových křivek, díky které se podařilo stanovit minimální inhibiční koncetraci 25 ?g/ml k testovaným kmenům E. coli. Difúzní testy prokázaly, že některé bakteriální kmeny P. larvae a M. luteus jsou citlivé vůči jelleinu I nebo jelleinu II, avšak necitlivé k apidaecinu Ia a abaecinu. Pro stanovení minimální inhibiční koncentrace jelleinu I a II vůči kmenům P. larvae je potřeba dále zoptimalizovat protokol kultivace v tekutém médiu. Získané výsledky poukazují na rozdílnou citlivost jednotlivých bakteriálních kmenů k antimikrobiálním peptidům, avšak rozdíly mohou být způsobeny i použitým kultivačním médiem nebo způsobem aplikace peptidů.Antimicrobial peptides are important part of humoral immunity of the animals. They occur across all organisms, including insect. In this group of animals was also isolated and characterized the largest amount of antimicrobial peptides. This thesis deals with the basic characteristic of antimicrobial peptides, their distribution into groups and mechanism of action. The thesis is focused on the antimicrobial peptides from honey bee (Apis mellifera), whose representatives (apidaecin Ia, abaecin, jelleine I, jelleine II) are also tested in the practical part of this thesis. The peptides are tested against the bacterial strains of Escherichia coli, Micrococcus luteus and also against Paenibacillus larvae, which is causative agent of American foulbrood. Cultivation conditions, use of suitable media and also application of the peptides to the bacterial culture was optimized for the testing by diffusion assay. The sensitivity of the E. coli strains against apidaecin Ia was determinated. Growth curves measuring method was applied to determine the minimal inhibitory concentration of apidaecin Ia against E. coli strains. Due to this method minimal inhibitory concetration of apidaecin Ia agaist E. coli strains was 25 ?g/ml. The results of diffusion tests showed that some of the P. larvae strains and M. luteus are sensitive against jellein I or jellein II, but these strains are not sensitive against apidaecin Ia and abaecin. To determinate the minimal inhibitory concentration of jellein I and jellein II against P. larvae strains is further optimalisation of cultivation in liquid medium required. The gained results point to different sensitivity of the bacterial strains to antimicrobial peptides. Both various composition of cultivation media and application of the peptides onto bacteria culture could cause different sensitivity of bacterial strains to antimicrobial peptides.
