Přítomnost UGT2B7 a VDR byla potvrzena téměř ve všech tkáních lidského těla. Jelikož VDR reguluje mnoho biotransformačních enzymů včetně UGT2B15 a UGT2B17, u kterých se společně s UGT2B7 předpokládá společný ancestrální předchůdce, je možné, že regulace exprese UGT2B7 je ovlivněna receptorem pro vitamín D. Pro ověření této hypotézy byl k aktivaci VDR použit kalcitriol a to ve třech fyziologicky relevantních koncentracích a sledovaný čas 24h byl použit pro detekci mRNA. Detekce proteinu byla stanovena za 48h. Téměř ve všech sledovaných tkáních (LS180, MDA MB231, CaCo-2, HepG2, MCF-7, A172, A549) byl prokázán funkční VDR, který po aktivaci kalcitriolem mnohonásobně zvýšil hladinu mRNA CYP24A1. Ve většině systémů, kde byl VDR aktivovaný kalcitriolem, došlo ke snížení hladiny mRNA UGT2B7. Nicméně se tento pokles neprojevil na hladině proteinu. Resveratrol, předpokládaný induktor exprese, měl v některých studovaných liniích represivní efekt (CaCo-2 a MCF-7). Sulforafan se jako induktor neprojevil ani v jedné ze studovaných tkání. Lze tedy říci, že kalcitriol negativně reguluje expresi mRNA UGT2B7 a ve střevní a prsní tkáni (LS180, CaCo-2, MDA MB 231, MCF-7), ale nijak neovlivňuje množství samotného proteinu (LS180 a HepG2).A presence of UGT2B7 and VDR was confirmed in almost all tissues of the human body. Because VDR regulates many biotransformation enzymes including UGT2B15 and UGT2B17 for which, along with UGT2B7, a common ancestral predecessor is assumed, it is possible that regulation of the expression of UGT2B7 is influenced by vitamin D receptor. To verify this hypothesis, calcitriol was used for activation of VDR, namely in three physiologically relevant concentrations and 24h observation was used to detect mRNA. Detection of protein was set in 48h. In almost all the observed tissues (LS180, MDA MB231, CaCo-2, HepG2, MCF-7, A172, A549) was demonstrated functional VDR, which activated by calcitriol greatly increased level of mRNA CYP24A1. In most systems, where VDR was activated by calcitriol, levels of mRNA UGT2B7 decreased. However, this decline was not reflected on the level of protein. Resveratrol, assumed inductor of expression, had a repressive effect in some studied lines (CaCo-2 and MCF-7). Sulforaphane as an inductor did not reflect in any of the studied tissues. Therefore we can say, that calcitriol negatively regulate expression of mRNA UGT2B7 and in intestinal and breast tissue (LS180, CaCo-2, MDA MB 231, MCF-7), but it does not influence an amount of the protein itself.
