Sekce Codonoprasum představuje v rámci rodu Allium jednu z taxonomicky nejkomplikovanějších sekcí. Jedná se o skupinu geofytních rostlin, jež svým rozšířením zasahuje od Středozemního moře po Kavkaz a která díky své rozmanitosti ve stupních ploidie vykazuje značnou diverzitu v morfologii, ekologii, reprodukčních systémech, či v karyologii. Teoretická část této bakalářské práce popisuje rovněž vznik polyploidních jedinců v populaci, zmíněn je i vliv polyploidie na rozmnožování, životaschopnost či celý genom jedince. Podrobně zpracovaný je i popis studovaných druhů rodu Allium sekce Codonoprasum s nástinem taxonomických přístupů k tomuto rodu. Nechybí ani přehled základních molekulárních metod, kterých se ke studiu fylogeneze vyšších rostlin, a zvláště pak polyploidních komplexů, využívá. V praktické části je následně popsána použitá metoda izolace genomové DNA a metoda AFLP, jež byla pro prvotní posouzení genotypové variability vybraných druhů rodu Allium sekce Codonoprasum z různých geografických populacích použita.The section Codonoprasum within the genus Allium represents, due to greatly morphological similarity of the species, high number of polyploids and polyploid complexes, one of the taxonomically most complicated sections. This is a group of geophytes, which are spread from the Mediterranean to the Caucasus. Diversity in ploidy levels demonstrates a considerable diversity in morphology, ecological tolerances, reproductive systems, and karyology. The theoretical part of this thesis describes different pathways of the polyploid formation, the effect of this process on reproduction, viability, and genome evolution of polyploids. There is also a detailed description of the studied species with an overview of approaches to the taxonomy of the genus Allium. The chapter number 3.6 is devoted to the basic molecular marker methods which are suitable for the phylogenetic studies of the higher plants, and especially of polyploids complexes. The practical part of this thesis consists of description of the genomic DNA isolation method and AFLP method. These methods were used for an initial assessment of genotypic variability of selected species of the genus Allium section Codonoprasum from different geographic populations.
