V teoretické části této bakalářské práce jsou uvedeny základní informace o proteomice, hmotnostní spektrometrii a přehled přístupů pro kvantifikaci proteinů. Dále je popsán proteinový hormon hepcidin-25 a jeho využití jako diagnostický biomarker onemocnění související s poruchou homeostázy železa. Experimentální část práce je zaměřena na vývoj a optimalizaci metody pro stanovení hladiny hepcidinu-25 v séru a moči pomocí kapalinové chromatografie ve spojení s tandemovou hmotnostní spektrometrií.In the theoretical part of this bachelor thesis are provided basic information about proteomics, mass spectrometry, and an overview of approaches for the quantification of proteins. It also describes a protein hormone hepcidin-25 and its use as a diagnostic biomarker for diseases associated with impairment iron homeostasis. The experimental part of this thesis is focused on the development and optimization of method for determining the levels of hepcidin-25 in human serum and urine by liquid chromatography coupled with tandem mass spectrometry.
