Při léčbě nádorových onemocnění se již řadu let využívají platinové komplexy. Tyto farmaka jsou označovány jako cytostatika a nejčastěji používaným lékem je cis-DDP. Mezi další se řadí oxaliplatina a karboplatina. Nevýhody spojené s klinickým použitím cis-DDP, jakými jsou např. nežádoucí vedlejší účinky na organismus pacienta a vrozená či opakovaným podáváním farmaka získaná rezistence některých nádorů, byly impulsem pro vznik nových protinádorových léčiv a následné detailnější studie molekulárních mechanismů jejich biologické účinnosti. Nové farmaka jsou převážně odvozena od cis-DDP nebo klinicky neúčinné trans-DDP. Náplní této práce bylo zjistit vliv velikosti aminoligandů trans- Pt(II) komplexů na interakci s DNA. Studovaným komplexem byl trans-[PtCl2(NH3)DEA], výsledky získané pomocí jednotlivých metod, byly porovnány s výsledky z dostupné literatury.Platinum complexes have been clinically used for many years to treat human malignancies. These pharmaceuticals are designated as cytostatics and most frequently used drug of this type is cis-DDP. Oxaliplatin and carboplatin belong to the next generation of platinum based cytostatics. Drawbacks associated with application of cis-DDP for treatment of cancer, e.g. severe side effects or the inherent and acquired resistance to this drug, gave an impuls to the design and synthesis of alternative transition metal-based anticancer drugs and consequently to studies focused on understanding details of the molecular mechanisms underlying the biological efficacy of these compounds. New pharmaceuticals are mainly derived from cis-DDP or clinically ineffective trans-DDP. The aim of this study was to determine the effect of size of aminoligands in trans-Pt (II) complexes on interaction of these complexes with DNA. Experimental studies were focused on trans-[PtCl2 (NH3) DEA], the experimental results were compared with results found in the literature.
