Pseudoperonospora cubensis je jedním z nejzávažnějších původců chorob rostlin čeledi Cucurbitaceae na světě. Výskyt tohoto patogenu z třídy Oomycetes je každoročně zaznamenán na území České republiky. Přirozená infekce je pozorována převážně na porostech Cucumis sativus a jen výjimečně na ostatních tykvovitých plodinách. Experimentální část této diplomové práce je zaměřena na determinaci variability virulence v populaci P. cubensis v České republice. Pro testování bylo použito celkem 118 izolátů pocházejících z různých krajů České republiky z let 2010-2012, převážně z porostů C. sativus, ale i z nových hostitelských druhů. Patogenní variabilita izolátů P. cubensis byla testována terčíkovou metodou v prostředí umělé inokulace s využitím diferenciačního souboru 12 genotypů z čeledi Cucurbitaceae (Lebeda a Widrlechner, 2003). Na testovaném souboru izolátů P. cubensis bylo determinováno 56 různých patotypů. Většina izolátů P. cubensis (75%) byla vysoce patogenní. Střední úroveň patogenity byla zaznamenána u 18% izolátů a nízká úroveň u 7% izolátů. Odolnost a náchylnost genotypů diferenciačního souboru vůči P. cubensis se ve sledovaném období lišila, a to především v roce 2011 v porovnání s ostatními roky. Detekce vysokého počtu patotypů P. cubensis zaznamenaná v letech 2010-2012 potvrdila dlouhodobý trend českých populací P. cubensis,a to jejich značnou proměnlivost v prostoru a čase.Pseudoperonospora cubensis is one of the most serious agents of disease for family Cucurbitaceae in a world. The presence of this pathogen in the Czech Republic observed every year. Natural infection is usually observed on Cucumis sativus and exceptionally on others cucurbit crops. Experimental part of this master thesis is focused on determination of virulence variation in population of P. cubensis in Czech Republic. Set of 118 samples originated in different regions of Czech Republic from years 2010-2012 were used for testing, mostly samples of Cucumis sativus, but also from new host species. Pathogenic variability of isolates of P. cubensis was tested by leaf-disc method in an artificial inoculation using differential set of 12 genotypes of the family Cucurbitaceae (Lebeda and Widrlechner, 2003). Fifty-six different pathotypes of P. cubensis were determined on tested set of isolates. Most isolates of P. cubensis (75%) was highly pathogenic. Medium level of pathogenicity was observed in 18% of the isolates and the low level of 7% of the isolates. Resistance and susceptibility of differential genotypes set against P. cubensis in the period of observation differed mainly in 2011 compared with other years.High number of pathotypes of P. cubensis detected in 2010-2012 confirmed the long-term trend of Czech´s P. cubensis populations and their considerable variability in space and time.
