Předložená práce je zaměřená na studium interakcí zástupců čeledi Asteraceae a padlí Golovinomyces cichoracearum. V rámci této diplomové práce byly makroskopicky studovány interakce mezi izoláty G. cichoracearum, které pocházely z různých druhů čeledi Asteraceae a mezi zástupci čeledi Asteraceae. Dále byl výzkum zaměřen na mikroskopické sledování vývoje G. cichoracearum na vybraných genotypech čeledi Asteraceae. Při studiu reakcí byla použita metoda cross-inokulačních testů, kdy byly makroskopicky hodnoceny nárůsty na jednotlivých listových discích a mikroskopicky sledován vývoj patogenu. Z realizovaných pozorování bylo zjištěno, že náchylné vůči patogenu G. cichoracearum byly především genotypy čeledi Asteraceae, které byly inokulovány izoláty patogenu pocházejícími z rostlin stejného druhu. Makroskopicky byly zjištěny léze padlí izolátu (GC/LS/1/11) z Lactuca serriola také na druzích Mycelis muralis (84/71), Hieracium murorum (89/378) a Tragopogon orientalis (49/239), z druhů Cichorium intybus (GC/CI/11) a C. endivia (GC/CE/11) na genotypu Lactuca serriola (LSE/57/15). Další výskyt padlí na testovaných rostlinách byl jen sporadický a výjimečný. Mikroskopickým sledováním vývoje byla zjištěna nejvyšší klíčivost konidií a sporulace opět u izolátů G. cichoracearum, které byly inokulovány na rostliny stejného druhu, jako byl původ těchto izolátů. Izoláty inokulované na genotypy odlišného druhu čeledi Asteraceae než byl původ izolátu, zastavily svůj vývoj ve fázi prvního konidiálního klíčního vlákna. Ze získaných výsledků je patrné, že se jedná o hostitelsky velmi specifický patogen, který rostl na rostlinách stejného druhu, odkud pocházel nebo na rostlinách taxonomicky blízkých původnímu hostitelskému taxonu.The presented work is focused on study of variability of the interactions between species of family Asteraceae and lettuce powdery mildew (Golovinomyces cichoracearum). In this thesis there were macroscopically sudied reactions between isolates of G. cichoracearum, which came from different genotypes of the Asteraceae family and between members of the family Asteraceae. Second goal of this research was microscopic observation of the development of G. cichoracearum on selected genotypes of the family Asteraceae. During the research the method of cross-inoculation tests was used, where the growth on leaf-discs was macroscopically evaluated and the development of the pathogen was microscopically monitored. There was found out, that mainly genotypes of family Asteraceae, which was inoculated by isolate of the pathogen originating from plant of the same species were susceptible to G. cichoracearum. Powdery mildew from Lactuca serriola (GC/LS/1/11) was macroscopically detected also on Mycelis muralis (84/71), Hieracium murorum (89/378) and Tragopogon orientalis (49/239). Powdery mildew from Cichorium intybus (GC/CI/11) and C. endivia (GC/CE/11) was detected on Lactuca serriola (LSE/57/15). Microscopic observation of the development of G.cichoracearum revaled that the most intensive germination of conidia and sporulation was found by these isolates which were inoculated on plants of the same species as origin of these isolates. Isolates, which was inoculated on different species of family Asteraceae stopped their development in stage of the first conidial germ tube. From these results is evident, that this host-pathogen interaction is a very specific and that isolates grew on the plants of the same species from which originated or on the plants which were taxonomically close to the original host taxon.
