Blumeria graminis f. sp. hordei, kauzální agens choroby ječmene označované jako padlí, je obligátně biotrofní patogenní houba, jejímuž studiu je vzhledem k rozsahu působených škod věnována značná pozornost. Výzkum tohoto modelového organismu v nedávné době zaznamenal výrazné úspěchy v podobě pozičního klonování prvních dvou genů avirulence a zejména projektu celogenomového sekvenování. Současný pokrok ve výzkumu zaměřeném na tento ekonomicky významný patogen shrnuje teoretická část této diplomové práce. Z uvedených poznatků pak vychází experimentální část, která se zaměřuje na odvození genotypovacího systému schopného rozlišit izoláty pocházející z geograficky omezeného území. S využitím dostupných sekvenčních dat byly navrženy a testovány markery založené na sekvencích mikrosatelitů a inzerčních místech transponovatelných elementů, které jsou velmi abundantní součástí genomu tohoto druhu. Vytvořený systém zahrnující 16 SSR, 14 SNP a 2 ISBP/RJM markery byl posléze aplikován na soubor 97 izolátů původem z České republiky, 50 australských izolátů a sbírku 11 izolátů reprezentujících světovou populaci padlí travního ječmene. Na základě použité sady markerů bylo dosaženo uspokojivého rozlišení studovaných izolátů, čímž se prokázalo, že navržený genotypovací systém představuje užitečný nástroj, který lze uplatnit při analýze diverzity, evolučních vztahů a prostorové i časové dynamiky izolátů na populační úrovni. V případě obohacení provedené analýzy o fenotypová data popisující virulence jednotlivých izolátů se otevírají další, atraktivní možnosti studia interakcí hostitele a patogenu v tomto modelovém systému.Fungus Blumeria graminis f. sp. hordei, a causal agent of the powdery mildew disease of barley, is an obligate biotrophic pathogen. Due to the significant yield losses it causes the pathogen attracts substantial attention of scientific community worldwide and has become a model organism. Recently, the research has resulted in a major breakthrough, besides the successful positional cloning of two avirulence genes, the whole genome sequencing project has been launched. The present-day progress in research carried out on this economically significant pathogen is summarized in the theoretical part of this study. Taking into account the information provided, the experimental part focuses on developing a genotyping system capable to characterize different isolates which originate from geographically limited area. Available sequence data have been used to design a set of molecular markers based on sequences of microsatellites and insertion sites of transposable elements which represent a very abundant part of the genome. The final genotyping system comprises of 16 SSR, 14 SNP and 2 ISBP/RJM markers which were subsequently applied on a set of 97 isolates originating from the Czech Republic, 50 Australian isolates and a collection of 11 isolates representing world population of barley powdery mildew. Marker set developed in this study has provided significant resolution of studied isolates and thus has proved to be a useful tool for analysis of diversity and evolutionary relationships as well as spatial and time dynamics of isolates on the population level. Being supplemented with additional phenotype data describing virulences present in individual isolates, this study might open new and attractive opportunities of studying host-pathogen relationship in this model system.
