Diplomová práce byla zaměřena na analýzu cukerné komponenty rekombinantního glykoproteinu gp120 metodou průtokové cytometrie s využitím fluorescenčně značených mikrosfér. Cukerná komponenta imobilizovaného gp120 byla analyzována pomocí tří lektinů s odlišnou glykanovou specifitou: AAL, GNL, PHA-L. Po nalezení optimální koncentrace lektinů byla studována změna reaktivity vybraných lektinů s gp120 před a po ošetřením enzymem PNGasa F.This thesis was focused on the analysis of oligosaccharide moiety of recombinant gp120 protein by flow cytometry using fluorescently labeled microspheres. A sugar component of immobilized gp120 was analyzed using three lectins with different glycan specificity: AAL GNL, PHA-L. After finding the optimal concentration of lectins,there were studied the changes of the reactivity of selected lectins with gp120 before and after treatment with the enzyme PNGasa F.
