Fytoplazmy jsou mikroorganismy bez pevné buněčné stěny způsobující onemocnění u stovek druhů rostlin. Na hostitelské organismy jsou přenášeny pomocí hmyzích vektorů. Taxonomicky jsou rozděleny především podle sekvence konzervovaného genu 16S r DNA, do 16 skupin. Práce se zabývá charakteristikou a identifikací fytoplazem předběžně zařazených do skupiny Bermuda grass white leaf (16SrXIV) infikujících plevele na Jižní Moravě. Fytoplazmy skupiny 16SrXIV se vyskytují téměř po celém světě, a infikují především jednoděložné rostliny, u kterých způsobují chlorózy a metlovitost. Hlavním zástupcem skupiny je Bermuda grass white leaf fytoplazma (Candidatus Phytoplasma cynodontis) parazitující hlavně na rostlinách troskutu prstnatého (Cynodon dactylon (L.) Pers.), vyskytující se nejvíce na asijském kontinentu. Molekulárně genetické analýzy dvou vzorků fytoplazem izolovaných z pcháče osetu Cirsium arvense (L.) Scop., vyloučily jejich příslušnost ke skupině fytoplazem Bermuda grass white leaf (16SrXIV) a prokázaly jejich fylogenetickou příbuznost s fytoplazmami skupiny Rice yellow dwarf (16SrXI). Detailní analýza sekvence 16S rDNA genu naznačila možnost, že se jedná o nový druh fytoplazmy. Pro potvrzení této skutečnosti je nezbytné analyzovat variabilitu dalších, a to neribozomálních genů.Phytoplasmas are wall-less microorganisms causing disease in hundreds of economically important plants.They are transmitted in the host organisms through the insect vectors. Taxonomically they are divided mainly accordning to the sequence of gene 16S rDNA into 16 groups. This work deals with characteristics and identification of phytoplasmas preliminary identified as a members of group Bermuda grass white leaf (16SrXIV) infecting weeds on the South Moravia. Phytoplasmas of group 16SrXIV can be found almost all over the world, in particular they infect monocots plants, to which they cause chlorosis and witches-broom. The major representative phytoplasma of group is Bermuda grass white leaf phytoplasma (Candidatus Phytoplasma cynodontis). It parasites mainly on the plants Bermuda grass (cynodon dactylon (L.) Pers.), which can be found mostly on the Asian continent. Molecular-genetic analysis of two samples of phytoplasmas isolated from Cirsium arvense (L.) Scop. Excluded their count among to the group Bermuda grass white leaf (16SrXIV) and have shown their phylogenetic relationship with group Rice yellow dwarf (16SrXI). Detailed analysis of sequence 16S rDNA gene indicated the possibility that it is a new species of phytoplasma. To confirme this fact, it is necessary to analyze variability of other nonribosomal genes.
