V teoretické části své bakalářské práce jsem se zabývala popisem řádu brodiví a jeho jednotlivých 5 čeledí. Největší pozornost jsem věnovala čeledi čápovití (Ciconidae), kam patří právě marabu africký. Dále jsem stručně popsala všechny dodnes nalezené polymorfní mikrosatelitové lokusy u ptáků z řádu brodiví. V poslední kapitole jsem se věnovala popisu repetitivních sekvencí a to konkrétně mikrosatelitům, jejich dělení, funkcím, nestabilitě, mutacím a hledání nových mikrosatelitových lokusů. Hledaní polymorfních mikrosatelitových lokusů pro čápa marabu jsem se věnovala v experimentální části. Využila jsem k tomu metodu cross-species PCR amplifikace a 187 párů primerů, původně navržených pro jiné druhy z řádů plameňáci (Phoenicopteriformes) a veslonozí (Pelecaniformes). Na konci experimentální části jsem nalezla 26 polymorfních mikrosatelitových lokusů. U nalezených polymorfních mikrosatelitů byla zoptimalizována teplota annealingu a délka elektroforetické separace. Z 26 páru primerů jich 18 bylo odvozeno od druhů z řádu veslonozí a zbylých 8 z řádu plameňáci. Počet alel se pohyboval od 2 do 7.In the theoretical part of my bachelor thesis I dealt with the description of the order Ciconiiformes and of its 5 families. I focused on the family Ciconidae, which includes Marabou Stork. I also briefly described all known polymorphic microsatellite loci of the order Ciconiiformes. In the last chapter I descripted repeat sequences and more specifically microsatellites, their division, features, instability, mutations and the search for new microsatellite loci. In the experimental part of my bachelor thesis I focused on the search of polymorphic microsatellite loci in Marabou Stork. I applied the method of cross-species PCR amplification, and I tested 187 primer pairs originally designed for the polymorphic microsatellites for birds belonging to orders of the flamingos (Phoenicopteriformes) and the pelicans (Pelecaniformes). Totally I found 26 of polymorphic microsatellite loci. For these polymophic microsatellite loci I optimized annealing temperature and a time of electrophoresis separation. Of the 26 pairs of primers 18 of them were derived from species of the order pelicans, and the remaining 8 of the order of flamingos. The number of allele ranged 2 to 7.
