Reaktivní formy dusíku (RNS) byly popsány jako signální molekuly v mnoha důležitých fyziologických a patologických procesech u vyšších rostlin. K RNS patří oxid dusný, oxid dusitý, oxid dusičný, peroxynitrit, S-nitrosothioly, S-nitrosoglutathion, a další. Diplomová práce se zabývá jejich popisem a následnou S-nitrosylací proteinů. S-nitrosylace proteinů je posttranslační modifikace bočního řetězce cysteinu, kdy se -NO skupina naváže na -SH skupinu cysteinu za vzniku S-nitrosylovaných proteinů nebo S-nitrosothiolů. K nejvýznamnějším z nich patří S-nitrosoglutathion, který je odbouráván S-nitrosoglutathionreduktasou (GSNOR), na kterou je tato práce zaměřena. GSNOR hraje v rostlinách významnou roli při detoxifikaci endogenního a exogenního formaldehydu a při regulaci hladiny S-nitrosothiolů. Tím rostliny chrání před nitrosativním stresem. V experimentální části práce byla zavedena metoda detekce aktivity rekombinantního enzymu GSNOR ze Solanum lycopersicon exprimovaného v E.coli na polyakrylamidových gelech po nativní elektroforéze. Dále byla aktivita tohoto enzymu stanovena spektrofotometrickými metodami. Tyto metody byly poté aplikovány při detekci aktivity u Arabidopsis thaliana a hrachu setého. Byla optimalizována metoda nativní elektroforézy pomocí elektronových akceptorů tvořících nerozpustné formazany. Byly pozorovány změny aktivit GSNOR u rostlin hrachu setého podrobených teplotnímu stresu. Toto prokazuje úlohu tohoto enzymu v obranné reakci rostlin v souvislosti s nitrosativním stresem.Reactive nitrogen species (RNS) are signalling molecules involved in many important physiological and pathological processes of higher plants. The RNS include, for example nitrous oxide, sulfur oxide, nitric oxide, peroxynitrite, S-nitrosothiols and S-nitrosoglutathione. S nitrotrosylated proteins are formed along with S-nitrosothiols by binding of -NO groups to -SH group of cysteine. One of the most significant S-nitrosylated peptids is S-nitrosoglutathione, which is degraded by S-nitrosoglutathione reductase (GSNOR). This enzyme ensures plants detoxification of endogenous and exogenous formaldehyde and regulates the level of S-nitrosothiols and thus protects plants against the nitrosative stress. The aim of this project was to optimize and apply new methods for detection of GSNOR enzyme activity. We used recombinant enzyme GSNOR from Solanum Lycopersicon expresed in E. coli for method optimalisation. Enzyme activity was detected after electrophoresis on native polyacrylamide gels using electron acceptors MTT, NBT and CTC. Furthermore, the enzyme activity was determined by spectrophotometric method. The optimized method was used for the detection of GSNOR activity in Arabidopsis thaliana and pea plants. Native electrophoresis method was optimized using electron acceptors forming insoluble formazans. Changes of GSNOR activity were observed in pea plants subjected to high temperature stress. This demonstrates the role of this enzyme in plant defence reactions in relation to the nitrosative stress.
