Transkripční faktory (TF) Ethylene Responsive Factors (ERF) regulují genovou expresi genů a zapojují se do růstů a vývoje rostliny. Tyto TF působí jako regulátory odezvy na biotické a abiotické stresy. V praktické části byla pomocí Agrobacterium tumefaciens provedena transformace nezralých zygotických embryí jarního ječmene odrůdy Golden Promise. Metoda CRISPR/Cas byla použita pro vyvolání mutace genu erf. Pomocí PCR byla ověřena přítomnost transgenů u rostlin v T0 generaci.The transcription factors (TF) Ethylene Responsive factors (ERF) regulate the gene expression and they engage in the growth and development of a plant. These TF act as controllers of the response to biotic and abiotic stress. In the practical part a transformation of the immature zygotic embryos of Hordeum of the Golden Promise species has been performer using the Agrobacterium tumefaciens. The metod CRISPR/Cas has been used in order to provoke the mutation of the gene erf. Via PCR the presence of transgenes in the plans of T0 generation has been verified.
