Tato diplomová práce se zabývá stanovením fenolických látek v destilátech. V teoretické části se zabývá stručnou charakterizací alkoholických nápojů se zaměřením na whisky. Dále se zabývá fenoly a jejich zdrojem původu ve whisky. Experimentální část je zaměřena na stanovení bromfenolů ve skotské whisky z ostrova Islay. Fenoly byly derivatizovány anhydridem kyseliny octové a extrahovány hexanem. Následně byly analyzovány plynovou chromatografií spojenou s hmotnostní spektrometrií (GC/MS). V této práci byly optimalizovány kroky derivatizace a parametry analýzy. Kvantifikace byla provedena metodou vnitřního standardu a metodou standardního přídavku. Ve vzorkách whisky byly nalezeny koncentrace bromfenolů v desítkách až stovkách ng/L, v případě 2-bromfenolu až v tisících ng/L.The main theme of this diploma's thesis is analysis of phenolic compounds in spirits. The theoretical part deals with a brief characterization of alcoholic beverages with a focus on whiskey. It also deals with phenols and their source of origin in whiskey. The experimental part is focused on the determination of bromophenols in Scotch whiskey from the island of Islay. Phenols have been derivatised by acetic anhydride and extracted using hexane. Consequently, the extracts have been analysed using gas chromatography mass spectrometry (GC/MS). Derivatisation steps and parameters of analysis were optimised in this thesis. The internal standard method and method of standard additions were used for a quantification. The concentration of bromophenols was found in tens to hundreds of ng/L and in case of 2-bromophenol in even up to thousands of ng/L in the whiskey sample.
