Diplomová práce je zaměřena na fukční vztah mezi methyltransferasami DNA a methylačními markery histonových modifikací u buněk mnohočetného myelomu. Byly testovány myelomové buněčné linie KMS- 12- PE, OPM-2 a U-266 po jejich ovlivnění dvěma typy demethylačních činidel - 5-aza-2´-deoxycytidin a 5- azacytidin. Přítomnost genů methyltransferas DNA - DNMT1, DNMT3A a DNMT3B v precipitovaných komplexech s protilátkami anti- H3K36me3/anti-H3K9me3 byla kvantifikována metodou RT-PCR a vyhodnocována statistickým Mann-Whitney testem.The diploma thesis is focused on the functional relationship between DNA methyltransferases and methylation markers of histone modifications in multiple myeloma cells. Myeloma cell lines KMS- 12- PE, OPM-2 and U266 were tested after their treatment with two types of demethylation agents - 5-aza-2´-deoxycytidine and 5- azacytidine. The presence of DNA methyltransferase genes - DNMT1, DNMT3A and DNMT3B in precipitated complexes with anti-H3K36me3/anti-H3K9me3 antibodies was quantified by RT-PCR and evaluated by statistical Mann-Whitney test.
