Diplomová práce se věnuje testování a charakterizaci vybraných polymorfních mikrosatelitů u třinácti nepříbuzných jedinců potápky roháče. V teoretické části je popsán řád potápky (Podicipediformes) včetně podrobného popisu potápky roháče, teoretické zpracování mikrosatelitů, cross-species PCR amplifikace a popis polymorfních a potencionálně polymorfních mikrosatelitů u potápky roháče. Experimentální část se věnuje testování třiceti dosud netestovaných mikrosatelitů u šesti nepříbuzných jedinců potápky roháče a genotypizace těch polymorfních společně s polymorfními mikrosatelity ze dvou bakalářských prací (Janušová, 2015; Filipová, 2016). Výsledkem bylo nalezení 42 polymorfních mikrosatelitů se dvěma až dvanácti alelami.The diploma thesis deals with testing and characterization of selected polymorphic microsatellite at thirteen unrelated individuals of the Crested Grebe. In the theoretical part it described Podicipediformes, including detailed description of Crested Grebe, theoretical treatment of microsatellites, cross-species PCR amplification and description of the polymorphic and potentially polymorphic microsatellites in Crested Grebe. In the experimental part deals with testing of thirty previously untested microsatellites in six unrelated individuals and genotyping of these polymorphic microsatellites together with polymorphic microsatellites from two bachelor thesis (Janušová, 2015; Filipová, 2016). It was found 42 polymorphic microsatellites with two to twelve alleles.
