Tato bakalářská práce je zaměřena na studium biologické aktivity konjugátů pohlavních hormonů s N-Boc-valinem in vitro. Tyto konjugáty, odvozené od testosteronu, estradiolu a 2-methoxyestradiolu, jsou zkoumány z důvodu potenciální antiproliferační aktivity. Teoretická část shrnuje poznatky o regulaci buněčného cyklu a programované buněčné smrti - apoptose. Pozornost je věnována také steroidním receptorům a jejich ligandům. V experimentální části byl studován vliv testovaných látek na nádorovou buněčnou linii HeLa, odvozenou od cervikálního karcinomu, a nádorovou linii MCF7, odvozenou od prsního adenokarcinomu. Pomocí metody western blottingu byla sledována exprese apoptotických markerů a steroidních receptorů. Následně byl metodou imunofluorescence pozorován vliv testovaných látek na translokaci steroidních receptorů z cytoplasmy do jádra.This bachelor thesis is focused on the study of biological activity of sex hormone conjugates with N-Boc-valine in vitro. These conjugates, derived from testosterone, estradiol and 2-methoxyestradiol, are investigated for their potential antiproliferative activity. The theoretical part summarizes the knowledge on cell cycle regulation and programmed cell death - apoptosis. Attention is also focused on steroid receptors and their ligands. In the experimental part, the effect of the tested substances on the HeLa tumor cell line, derived from cervical carcinoma, and the MCF7 tumour line, derived from breast adenocarcinoma, was studied. The expression of apoptotic markers and steroid receptors was monitored by western blotting. Subsequently, the effect of the tested substances on the translocation of steroid receptors from the cytoplasm to the nucleus was observed by immunofluorescence.
