Pšenice je základem stravy pro 40 % světové populace a vyrábí se z ní většina nezbytných potravin. Nárust zalidnění ovšem způsobuje neustálý tlak na zvýšení její produkce. Z toho důvodu je nutné šlechtění nových kultivarů, které by se mohly lépe adaptovat na měnící se podmínky prostředí. Správné načasování doby kvetení je důležité pro reprodukční úspěch rostliny. Jedna z genetických drah, která kvetení ovlivňuje, je vernalizační dráha. Tato bakalářská práce se zaměřuje na sekvenční analýzu a hledání nových alel u nejdůležitějšího vernalizačního genu VRN-1 u 93 genotypů tetraploidní pšenice plané dvouznky (Triticum dicoccoides). V rámci teoretické části práce jsou popsány hlavní molekulární dráhy ovlivňující kvetení u modelového organismu Arabidopsis thaliana a pšenice, se zaměřením na vernalizaci. Dále teoretická část pojednává o vybraných metodách sekvenování. V experimentální části práce byly sekvenovány VRN-A1 a VRN-B1 promotory u celkem 93 genotypů T. dicoccoides z oblasti Úrodného půlměsíce, které byly následně spojeny se sekvencemi celých genů za účelem analýzy sekvenční variability. V rámci VRN-A1 i VRN-B1 genů bylo nalezeno velké množství mutací, včetně mutací v exonech. Celkem bylo nalezeno 12 nepopsaných mutací u VRN-A1 (jedna delece v oblasti promotoru, pět SNPs v exonech a šest indelů v intronech) a 21 u VRN-B1 (tři indely v promotoru, sedm SNPs v exonech a 11 indelů v intronech), jejichž vliv na kvetení je třeba charakterizovat a následně vybrat ty alely, které mohou být použity v rámci šlechtění.Wheat is one of the world's main food sources (40 %) and is used to make most of the necessary foodstuff. However, the increase of population causes constant pressure to increase its production. For this reason, it is important to breed new cultivars that would be more resistant to deteriorating environmental conditions and could flower in shorter time. The timing of flowering is essential to the reproductive success of the plant. One of the genetic pathways that affect this flowering period is vernalization. This bachelor thesis is focused on the sequence analysis of VRN-1 genes, which play a crucial role in the initiation of flowering, in selected cultivars of wild emmer wheat (Triticum dicoccoides) and the search for new alleles that could be used in breeding. The theoretical part of the work describes the main molecular pathways that affect flowering in the model organism Arabidopsis thaliana and wheat, with the main focus on vernalization. The theoretical part also includes selected sequencing methods. In the experimental part of the work, VRN-A1 and VRN-B1 promoters were sequenced in a total of 93 genotypes of T. dicoccoides from the Fertile Crescent area. The promoter sequences were subsequently merged with the sequences of whole genes in order to analyze the sequence variability. A large number of mutations, including mutations in exons, have been found within both VRN-A1 and VRN-B1 genes. There were a number of SNPs (single nucleotide polymorphisms) found in the introns, as well as larger insertions and deletions. A total of 12 undescribed mutations were found in VRN-A1 (one deletion in the promoter region, five SNPs in exons and six indels in introns) and 21 in VRN-B1 (three indels in the promoter region, seven SNPs in exons and 11 indels in introns). The effect of these mutations on flowering is to be characterized to choose alleles that can be used in breeding.
