Tato bakalářská práce se zabývá stanovením endogenních hladin cytokininů v Arabidopsis thaliana v průběhu dne. Tyto rostlinné hormony jsou díky procesům, které ovlivňují, pro růst a vývoj rostliny zásadní a jejich koncentrační hladiny se cyklicky mění v závislosti na denní době. Rostliny byly pěstovány ve fytotronu v kontrolovaných podmínkách s pevně stanovenou denní a noční fází a desetidenní semenáčky byly sklízeny v předem stanovených intervalech během jednoho dne. Vzorky byly následně purifikovány pomocí StageTip metody a cytokininy a jejich metabolity v nich byly stanoveny pomocí ultra-vysokoúčinné kapalinové chromatografie v zapojení s tandemovou hmotnostní spektrometrií.This bachelor thesis deals with the determination of endogenous levels of cytokinins in Arabidopsis thaliana during the day. Due to the processes they affect, these plant hormones are necessary for growth and development of plants and their concentrations change cyclically depending on the time of the day. Plants for the experimental part were grown in phytotron with controlled conditions with a set day and night phase and the ten-day-old seedlings were reaped in set intervals during one day. Samples were purified using the StageTip method and cytokinins and their metabolites were quantified using ultra-high performance liquid chromatography with tandem mass spectrometry.
