Cílem první části dizertační práce je podat ucelený přehled problematiky přípravy a purifikace rekombinantních proteinů a možností jejich využití v diagnostice a terapii lidských chorob. Jsou zde popsány technologické postupy izolace cílového genu, tvorby rekombinantního expresního vektoru, vlastnosti jednotlivých expresních systémů a metodiky purifikace rekombinantních proteinů. Druhá část práce obsahuje shrnutí řešené problematiky. Při přípavě rekombinantních proteinů jsem se zabýval třemi dílčími tématy. Prvním tématem byla příprava povrchových antigenů spirochety Borrelia burgdorferi a testování jejich imunogenních vlastností na myším modelu. Druhým tématem byla purifikace rekombinantního proteinu Hsp90 kvasinky Candida albicans a testování jeho využitelnosti jako antigenu pro přípravu preventivní vakcíny proti kandidóze. Posledním tématem byla exprese a purifikace rekombinantního myšího proteinu Hsp70 pro vakcinační experimenty. Výsledky práce na uvedených tématech vedly k publikace 8 prací v recenzovaných časopisech. Výtisky prací tvoří třetí část dizertační práce.The aim of the first part of the PhD thesis is to bring the comprehensive overview of preparation and purification of recombinant proteins and their utilization in diagnosis and therapy of human deseases. Here are described technological procedures of target gene isolation, construction of recombinant vectors, properties of expression systems and methods of recombinant protein purification. The second part contains summary of experimental work on recombinant protein production which is divided in three subjects. First, the production of surface antigens from spirochete Borrelia burgdorferi and testing their immunogenic properties on mouse model. Second, the purification of recombinant protein Hsp90 from yeast Candida albicans and testing of its applicability as antigen for preparation of preventive vaccine against candidiosis. Third, the expression and purification of recombinant mouse Hsp70 protein for vaccination experiments. The results of the investigation were published in eight papers of which five are in journals with impact factor and rest in per-reviewed journals. Copies of these papers are attached in the third part of the thesis.
