Již dlouhou dobu jsou známy pozitivní účinky cytokininů na oddálení senescence rostlin. Zajímavé ovšem je, že tomu tak je pouze do určité koncentrace aplikovaných cytokininů. Cílem této práce bylo zkoumat vliv exogenně aplikovaných cytokininů a světla na senescenci listů ječmene jarního se sníženým obsahem těchto hormonů. Senescenční testy byly provedeny s oddělenými listy transgenní linie ječmene s overexpresí proteinu CKX1 a s WT rostlinami jako kontrolou. Byla použita řada experimentálních metod, jako měření obsahu cytokininů, analýza obsahu chlorofylu analyticky i pomocí chlorofylmetru či stanovení aktivity fotosyntetického aparátu. Byl prokázán vliv zvyšující se koncentrace cytokininů na snížení degradace chlorofylu za současného snížení fotosyntetické aktivity fotosystému II.It is known for a long time that cytokinins have positive effects on delay of plant senescence. Interestingly, this is true only up to certain concentration of applied cytokinins. The aim of this thesis was to investigate the effect of exogenously applied cytokinins and light on senescence of spring barely leaves with decreased levels of those hormones. Senescence assays were performer on detached leaves of transgenic barley with CKX1 protein overexpression and WT plants as a control. Several experimental methods were used, such as estimation of cytokinins content, analysis of chlorophyll content analytically as well as using chlorophyllmeter, and measurement of activity of photosynthetic apparatus. The effect of increasing cytokinin concentration on the reduction of chlorophyll degradation has been demonstrated, while the photosynthetic activity of photosystem II was reduced.
