Esenciální oleje jsou koncentrované hydrofobní kapaliny obsahující nejen sekundární metabolity rostlin, které díky svým rozmanitým vlastnostem a biologické aktivitě našly využití v mnoha odvětvích včetně kulinářství. Je známo, že některé esenciální oleje, které se používají právě v kulinářství, jsou schopné interagovat s aryluhlovodíkovým receptorem, který se účastní regulace mnoha fyziologických procesů v organismu. Vzhledem k tomu, že esenciální oleje jsou mnohasložkové směsi, je důležité identifikovat AhR-aktivní komponenty. Bakalářská práce je zaměřena na studium vlivu vybraných složek esenciálních olejů na transkripční aktivitu aryluhlovodíkového receptoru. K testování byly vybrány látky (1S)-(+)-3-caren, terpinolen, terpinen-4-ol, (+)-aromadendren, myristicin, cuminol, methylsalicylát, beta-myrcen, borneol a camphen. Pro určení rozmezí netoxických koncentrací testovaných látek byl použit MTT test. Vliv těchto látek na transkripční aktivitu AhR byl zkoumán za využití metody Reporter Gene Assay ve dvou módech: agonistickém a antagonistickém. Z testovaných látek byly jako parciální agonisté AhR hodnoceny myristicin a cuminol. Antagonistická aktivita byla pozorována v případě myristicinu při použití v kombinaci s modelovým ligandem TCDD, ovšem ale v přítomnosti modelového ligandu FICZ pokles indukce pozorován nebyl. Ovšem pro lepší porozumění probíhajícím interakcím a jejich případné využití by bylo potřeba dalšího výzkumu.Essential oils are concentrated hydrophobic liquids containing not only secondary metabolites of plants that, due to their diverse properties and biological activity, have found use in many sectors, including culinary. It is known, that some of the essential oils of culinary herbs and spices, can interact with the aryl hydrocarbon receptor, which is involved in regulation of many physiological processes of the organism. Since essential oils are multicomponent mixtures, it is important to identify AhR-active constituents. The bachelor thesis is focused on effects of selected components of essential oils on transcriptional activity of aryl hydrocarbon receptor. Compounds selected for testing were (1S)-(+)-3-carene, terpinolene, terpinen-4-ol, (+)-aromadendrene, myristicin, cuminol, methyl salicylate, beta-myrcene, borneol and camphene. For determination of non-toxic concentrations of tested substances, MTT test was used. The effect of selected components on transcriptional activity of AhR was monitored using Reporter Gene Assay in two modes: agonist and antagonist mode. Of the tested substances myristicin and cuminol were identified as partial agonists of AhR. An antagonistic effect on AhR was observed in case of myristicin when used in combination with the model ligand TCDD, but in the presence of the model ligand FICZ, no induction decrease was observed. However, for better understanding of ongoing interactions and their possible use, further research would be needed.
