Studovala jsem biodiverzitu čeledi Lycidae (Coleoptera), konkrétně trichaliních rodů subtribu Metriorrhynchina, v tropické australsko-orientální oblasti. Pro experimentální část práce byla použita datová matice nově sekvenovaných fragmentů mitochondriálního markeru cox1, ke které byly dodány mitochondriální markery rrnL, nad5 a jaderné fragmenty LSU rRNA a SSU rRNA. Z takto konkatenovaného datového souboru byl metodou maximum likelihood vytvořen fylogenetický strom, jehož výsledky byly interpretovány s ohledem na morfologickou diverzitu. Delimitační metoda bPTP byla použita pro srovnání počtu druhů v analýze a formálně pojmenovaných druhů v rámci taxonomických studií. Výsledky prokazují druhovou radiaci trichaliních rodů hlavně v oblasti Moluk (rod Flabellotrichalus) a Nové Guinei (rody Eniclases a Microtrichalus). Nově získaná sekvence fragmentu genu cox1 pro rod Schizotrichalus z Moluk vyřešila fylogenetickou pozici této skupiny. Fylogeneze navíc odráží tektonickou historii oblasti a potvrzuje dřívější propojení severní části Austrálie a Nové Guinei jako oblasti, které jsou centrem radiace terminální linie subtribu Metriorrhynchina, což je v souladu s předešlými studiemi.Here, I studied the diversity of the elateroid family net-winged beetles (Coleoptera: Lycidae), namely the group of the trichaline genera, from the tropical part of the Australian and Oriental regions and from the transitional area between them, called the Wallacea. The laboratory experiments were conducted to produce the new sequences of cox1 mitochondrial DNA fragment. Newly produced data were merged with earlier published rrnL and nad5 mtDNA fragments and nuclear LSU rRNA a SSU rRNA fragments. The concatenated dataset was analysed using the maximum likelihood criterion and I obtained the robustly supported phylogenetic hypothesis on the relationships among main lineages of trichaline beetles. I used the bPTP delineation method for the estimation of diversity and compared the number of identified species with the formally described diversity in the region. The results suggest rapid diversification in the area of New Guinea (Eniclases and Microtrichalus) and the Moluccas (Flabellotrichalus). The newly produced data solved the phylogenetic position of the genus Schizotrichalus from the Moluccas. The phylogeny reflects the tectonic history of the region. I have found high similarity between faunas of Northern Australia and New Guinea which had been connected during Quaternary Glacial Maxima. These regions are the cradle of the diversity of Metriorrhynchine beetles as has been earlier suggested by the studies dealing with the relatives of the trichaline genera.
