Tato práce se zabývá vlivem látky grafen oxidu na transkripční aktivitu jaderného receptoru pro vitamín D (VDR). V teoretické části bakalářské práce je nastíněn současný stav dané problematiky. Je zde popsána funkce vitamínu D, jeho izoformy a význam pro lidský organismus. Další kapitola je věnována rodině jaderných receptorů a nalezneme zde podrobnější informace o studovaném jaderném receptoru, čili receptoru pro vitamín D. Následující kapitola se věnuje nanomateriálům, grafenu a samotnému grafen oxidu, jeho struktuře a významu. Experimentální část je rozdělena na dva soubory experimentů. V první části bylo pomocí MTT testu zjišťováno cytotoxické působení částic grafen oxidu na VDR-responzivní buněčnou linii. Výsledky dokázaly, že žádná z použitých koncentrací grafen oxidu nemá na VDR-responzivní buněčnou linii toxické účinky a tudíž mohou být pro další část experimentů použit všechny testované koncentrace. Ve druhé části byla zjišťována schopnost těchto částic aktivovat nebo inhibovat transkripční aktivitu VDR metodou Gene reporter assay. Aktivita VDR byla stanovována pomocí reportérového genu pro luciferázu, který je pod kontrolou VDR responzivních elementů.This bachelor thesis deals with influence of graphene oxide nano-platelets on the transcriptional activity of the nuclear receptor for vitamin D (VDR). The theoretical part of this thesis defines the current statement of this problematics. Vitamin D functions are described, also its isoforms and its importance for human organism. The next chapter is dedicated to the family of nuclear receptors and we can find here also more information about the studied nuclear receptor, the vitamin D receptor. The following chapter deals with nanomaterials, graphene and graphene oxide itself, its structure and significance. The experimental part is divided into two sets of experiments. In the first part, the cytotoxic effect of the graphene oxide nano-platelets on the VDR-responsive cell line was detected by the MTT test. The results demonstrated that none of used concentrations of graphene oxide had toxic effect on the VDR-responsive cell line and therefore all tested concentrations could be used for all other experiments. In the second part, it was determined the ability of these particles to activate or inhibit the transcriptional activity of VDR using the Gene reporter assay. VDR activity was determined using a reporter gene for luciferase, which is under the control of VDR responsive elements.
