Bakalářská práce napomáhá objasnit, jakým způsobem jsou cytokininy transportovány přes membránu vakuoly. V teoretické části práce byla zpracována literární rešerše o cytokininech, jejich struktuře a biologické aktivitě v rostlinách. Dále byl vypracován přehled týkající se jejich metabolismu a transportu. Součástí experimentální části byla genotypizace vybraných mutantních linií Arabidopsis thaliana s deletovanými geny domnělých cytokininových přenašečů přes vakuolární membránu pomocí metody PCR. Získané ověřené homozygotní rostliny budou dále použity na fenotypovou analýzu.The bachelor thesis helps to clarify how are cytokinins transported through the vacuolar membrane. A literature review about cytokinins, their structure and biological activity in plants was written in theoretical part of the thesis. Further, summary relating to their metabolism and transport was made. In the experimental part selected mutant lines Arabidopsis thaliana with deleted genes, which are hypothetically involved in the transport of cytokinins through the vacuolar membrane were genotyped. Verified homozygous mutant plants will be further subjected to detailed phenotypic analysis.
