Tato bakalářská práce se zabývá problematikou alogenních transplantací hematopoetických kmenových buněk (HSC) u pacientů s hematoonkologickými onemocněními a možnostmi sledování hladiny buněčného chimerismu metodami molekulárně biologickými aj. u pacientů, kteří alogenní transplantaci HSC podstoupili. Teoretická část shrnuje základní fakta o kmenových buňkách s důrazem na HSC a krvetvorbu. Také se věnuje problematice transplantací HSC a laboratorním metodám vyžívaným při sledování hladiny dárcovského chimerismu po alogenních transplantacích HSC. Experimentální část zahrnuje vyšetření předtransplantačních a následně i potransplantačních vzorků u vybraných pacientů pomocí STR-PCR a fragmentační analýzy. Pomocí této metody byla provedena genotypizace 4 pacientů a jejich dárců a analyzováno 19 potransplantačních vzorků, u nichž bylo sledováno postupné přihojování štěpu, remise a u 2 pacientů i relaps onemocnění.This bachelor thesis is focused on allogeneic transplantations of hematopoietic stem cells (HSCT) in patients suffering from hematologic malignancies. It describes available methods for chimerism monitoring by molecular methods in case of patients, who has undergone the HSC transplantation. The theoretical part summarizes the information about stem cells - hematopoietic stem cells in particular, and hematopoiesis. Furthermore, it deals with the topic of HSC transplantations and the methods used for monitoring of donor chimerism levels after the allogeneic HSCTs. The experimental part presents pre-transplant and post-transplant analyses of samples from 4 chosen patients. Analyses consist of genotypizations of the patients and their donors and post-transplant evaluation of 19 samples. The method used in this case is STR-PCR followed by fragment analysis performed on genetic analyser ABI 3100. The engraftment, as well as remision and relaps have been detected.
