Bakalářská práce se věnuje testování mikrosatelitů z řádu trubkonosí na polymorfismus u šesti nepříbuzných jedinců potápky roháče. V teoretické části je popsán řád potápky (Podicipediformes) včetně podrobného popisu potápky roháče, teoretické zpracování mikrosatelitů, cross-species PCR amplifikace a popis doposud objevených mikrosatelitů izolovaných od zástupců řádu trubkonosí. Experimentální část se věnuje testování 213 párů primerů na cross-species PCR amplifikaci u potápky roháče. Pomocí této metody bylo nalezeno u tohoto druhu 20 polymorfních mikrosatelitů se dvěma až osmi alelami.Bachelor thesis deals with testing microsatellites for polymorphism in six unrelated individuals of Crested Grebe. In the theoretical part it described Podicipediformes, including detailed description of Crested Grebe, theoretical treatment of microsatellites, cross-species PCR amplification and description of previously discovered microsatellite isolated from representatives of Procellariiformes. In the experimental part deals with testing cross-species PCR amplification of 213 primer pairs in Crested Grebe. Using the method it found a total number of 20 polymorphic microsatellites with two to eight alleles.
