Práce se zabývá studiem vybraných imunogenetických faktorů u alogenních transplantací kmenových krvetvorných buněk. Základem dizertační práce je osm publikací, které byly publikovány (některé i citovány) v mezinárodních časopisech s impakt faktorem a jsou v práci vloženy jako separátní výtisky v originálním anglickém znění. Práce je členěna do tří základních oddílů. První část, literární přehled, shrnuje problematiku transplantací kmenových krvetvorných buněk, popisuje možné komplikace tohoto terapeutického přístupu i faktory, které rozvoj zmíněných komplikací ovlivňují. Dále se literární přehled zaměřuje na charakteristiku vybraných imunogenetických faktorů studovaných v souvislosti s transplantacemi kmenových krvetvorných buněk a krátce pojednává i o metodách jejich studia. Druhá část vymezuje cíle práce, popisuje studované soubory a použité metodické přístupy. Ve třetí části jsou pak předkládány výsledky práce, která se věnovala studiu genetických faktorů ovlivňujících výsledek alogenních transplantací kmenových krvetvorných buněk. V obou výsledkových podkapitolách experimentální části jsou shrnuty cíle provedených studií a použité metodické přístupy. Nejvýznamnější nálezy z vlastních prací jsou shrnuty a diskutovány s ohledem na aktuální úroveň znalostí.The thesis is devoted to the study of selected immunogenetic factors of the allogeneic haematopoietic stem cell transplantation (AHSCT). It is based on the eight manuscripts published in the international impacted journals and inserted in the thesis as separate printouts in original English version. The thesis is divided into the three basic sections. The first section, literary overview, summarizes the AHSCT issue, describes possible complications of this therapeutic approach and factors which may have an influence to their development. The overview also focuses on the characteristics of selected immunogenetic factors studied in the context of AHSCT and shortly deals with the methods of their study. The second section of the thesis defines its aim, describes study groups and methodological approaches which were used in the individual studies. The results of the study of the genetic factors influencing the AHSCT outcome are presented in the third section, experimental part of the thesis. The most significant findings of the research are summarised and discussed with respect to the current knowledge.
