Náplní práce je souhrnná charakterizace principiálních chromatografických technik. V teoretické části je důraz kladen na vysvětlení pojmů, principů a procesů základních chromatografických metod. Pozornost je také soustředěna na jednotlivé konkrétní techniky, zejména na kapalinovou chromatografii, a to gelovou permeační chromatografii a iontově výměnnou chromatografii. V experimentální části práce je přiblížena technika a metodika separace a purifikace lidského polymerního imunoglobulinu IgA pomocí gelové permeační chromatografie a iontově výměnné chromatografie od ostatních proteinů krevní plasmy.The purpose of the thesis is an overall characterization of principal chromatographic techniques. The theoretical part emphasizes the explanation of terms, principals and processes of basic chromatographic methods. Special attention is payed to particular techniques, especially to liquid chromatography, namely gel permeation chromatography and ion exchange chromatography. The experimental part examines more closely the technique and methodology of separation and purification of human polymeric imunoglobulin IgA from the other proteins of blood plasma by means of gel permeation chromatography and ion exchange chromatography.
